چکیده
A role for the orexin-1 receptor (OX1R) in emotional behavior is emerging. The present study characterizes the brain-penetrant OX1R antagonist, compound 56 [N-({3-[(3-ethoxy-6-methylpyridin2-yl)carbonyl]-3-azabicyclo[4.1.0]hept-4-yl}methyl)-5-(trifluoromethyl) pyrimidin-2-amine]. Receptor binding studies demonstrated that compound 56 crossed the blood-brain barrier and occupied OX1Rs in the rat brain. While compound 56 did not alter sleep in wild-type mice, its administration in OX2R knockout mice selectively promoted rapid eye movement sleep, demonstrating target engagement. In a rat model of stress induced by cage exchange, the OX1R antagonist prevented prolongation of sleep onset without affecting sleep duration. In a rat model of panic vulnerability (involving disinhibition of the perifornical OX region), the compound attenuated sodium lactate–induced panic-like behaviors without altering baseline locomotor or autonomic activity. In conclusion, OX1R antagonism represents a novel therapeutic strategy for the treatment of various psychiatric disorders associated with stress.

Administration of orexin receptor 1 antagonist into the rostral ventromedial medulla increased swim stress-induced antinociception in rat
Objective(s): Intracerebroventricular injection of orexin-A (hypocretin-1) antagonist has been shown to inhibit stress-induced analgesia. However the locations of central sites that may mediate these effects have not been totally demonstrated. This study was performed to investigate the role of rostral ventromedial medulla (RVM) orexin receptor 1 in stress-induced analgesia (SIA). Materials and...
متن کاملA selective orexin-1 receptor antagonist attenuates stress-induced hyperarousal without hypnotic effects.
Orexins (OXs) are peptides produced by perifornical (PeF) and lateral hypothalamic neurons that exert a prominent role in arousal-related processes, including stress. A critical role for the orexin-1 receptor (OX1R) in complex emotional behavior is emerging, such as overactivation of the OX1R pathway being associated with panic or anxiety states. Here we characterize a brain-penetrant, selectiv...
متن کاملAssesment of orexin receptor 1 in stress attenuated nociceptive behaviours in formalin test
Introduction: It is known that acute and chronic stress induce hormonal and neuronal changes which affecting both pain threshold and nociceptive behaviours. Orexin plays an important role in modulation of pain and stress. Considering pain modulation during stress and the role of orexin in pain and stress, orexin might be involved in pain modulation during stress. We evaluated the involvement ...
متن کاملSelective Inhibition of Orexin-2 Receptors Prevents Stress-Induced ACTH Release in Mice
Orexins peptides exert a prominent role in arousal-related processes including stress responding, by activating orexin-1 (OX1R) and orexin-2 (OX2R) receptors located widely throughout the brain. Stress or orexin administration stimulates hyperarousal, adrenocorticotropic hormone (ACTH) and corticosterone release, and selective OX1R blockade can attenuate several stress-induced behavioral and ca...
متن کاملRole of Orexin-1 Receptor Within the Ventral Tegmental Area in Mediating Stress- and Morphine Priming-induced Reinstatement of Conditioned Place Preference in Rats
Introduction: Orexin-containing neurons exist in the lateral hypothalamic region, sending their projections toward mesolimbic regions such as the Ventral Tegmental Area (VTA). Methods: In the current study, a Reinstatement model is used to examine the effects of intra-VTA administration of SB334867 as an Orexin-1 Receptor (OX1R) antagonist on drug priming- and Forced Swim Stress (FSS)-induced ...
